چکیده

Cellulase activity was measured in soybean (Glycine max) leaf abscission zones, flower abscission zones, pod abscission zones, apical buds, and adventitious rooting hypocotyls. Immunoprecipitation data showed that a cellulase immunologically similar to the bean abscission cellulase (isoelectric point 9.5) is present in soybean leaf, flower, and pod abscission zones, but is not present in soybean apical buds or rooting hypocotyls. cDNA and genomic clones for two different soybean genes were identified and show sequence similarity with the bean abscission cellulase clone pBAC10. The cDNA clone pSAC1, isolated from a soybean abscission cDNA library, hybridized to transcripts in soybean leaf, flower, and pod abscission zones. Although ethylene has been shown to play a role in the increase in cellulase activity associated with both abscission and adventitious root initiation, no signal was seen for hybridization of the soybean abscission cellulase clone, pSAC1, to RNA from soybean adventitious rooting hypocotyls. In addition, no soybean abscission cellulase transcripts were detected in apical buds. Transcripts for a second soybean cellulase gene (SC2) were not detected in any of the tissues surveyed.

Gene expression profiles for cell wall-modifying proteins associated with soybean cyst nematode infection, petiole abscission, root tips, flowers, apical buds, and leaves.

Changes in transcript accumulation for cell wall-modifying proteins were examined in excised soybean root pieces colonized by soybean cyst nematodes (SCN), Heterodera glycines, using RT-PCR and soybean Affymetrix GeneChips. Sequence-specific PCR primer pairs were prepared from sequence data for core sequences in the GenBank soybean database and consensus sequences derived from the assembly of s...

GROWTH SUBSTANCE AND THE DEVELOPMENT OF CROWN GALLi

Advances made recently in the study of plant growth substances and their function in normal growth have raised the question of their possible role in atypical and pathological growth and specifically in the gall development induced by Phytomonas tumefaciens (Smith and Town.) Bergey et al. A suggestion of their presence was first given by the marked epinasty of leaf petioles in plants inoculated...

The synthetic growth regulant DPX 1840 (3,3a-dihydro-2-(p-methoxyphenyl)-8H-pyrazolo[5, 1-a]isoindol-8-one) induced callus growth and subsequent tissue differentiation on cut surfaces of decapitated Xanthi-nc tobacco plants (Nicotiana tabacum). Callus formation and organogenesis induced by DPX 1840 depended on the presence of leaves. The adventitious meristems developed into either vegetative o...
